Objectifs
L’équipe ECI a pour objectif de comprendre l’impact des contaminants chimiques d’origine anthropique sur les assemblages biologiques, dans un contexte de réalisme écologique, en considérant notamment les interactions des contaminants entre eux et avec d’autres facteurs de stress.
Pour cela, l’équipe ECI s’appuiera sur les outils développés lors des contrats précédents, notamment en matière de biomarqueurs de (géno)toxicité. L’approche intégrative passera par l’expérimentation en micro-/mésocosmes, dans le but d’appréhender en conditions contrôlées l’impact des contaminants sur des assemblages plurispécifiques, avec l’aide de la modélisation utilisée à des fins prédictives et cognitives. L’équipe ECI aura à cœur d’appuyer sa démarche sur des chantiers d’étudein natura, concernés par des problématiques concrètes de contamination (et concentrant par ailleurs des moyens financiers et humains). La Zone Atelier Pyrénées-Garonne offrira localement cette possibilité. Les travaux initiés en Camargue, concernant l’impact des contaminations environnementales sur les herbiers de zostères, seront poursuivis. L’équipe ECI sera par ailleurs impliquée, au sein d’un consortium franco-équatorien, dans l’étude de l’impact de pollutions par les hydrocarbures et métaux associés sur des cours d’eau amazoniens.
L’originalité de la démarche de l’équipe ECI réside dans la mise en œuvre d’approches complémentaires à différents niveaux d’organisation biologiques pour progresser dans la compréhension des impacts environnementaux liés aux contaminations chimiques.
Objectifs Scientifiques – Questions
1) Quels sont les mécanismes conditionnant la sensibilité des organismes au stress chimique ?
De nombreuses questions demeurent concernant l’effet combiné des contaminants en mélange, ou en interaction avec d’autres stress environnementaux. Par ailleurs, Les mécanismes d’action de contaminants émergents, tels que certains nanomatériaux manufacturés, sont encore loin d’être totalement élucidés.
L’équipe ECI dispose des compétences et des outils pour appréhender les mécanismes de toxicité liés notamment au stress oxydant, aux dommages causés au matériel génétique, ou encore aux altérations du métabolisme énergétique des organismes. Une attention particulière sera accordée à ce dernier point, dans la mesure où le métabolisme énergétique joue un rôle central dans la survie des organismes et se trouve fortement sollicité en condition de stress pour la mise en place des systèmes de défense. Le rapport coût / bénéfice de la réponse plastique aux stress peut ainsi conditionner la valeur adaptative des individus et la dynamique des populations. Nous utiliserons des outils moléculaires tels que la protéomique et la métabolomique afin d’évaluer les effets sur les organismes de situations de stress complexes (variations de température, cocktails de contaminants…).
Une attention particulière sera portée à la variabilité intraspécifique de la réponse des organismes aux contaminants, à ce jour encore peu étudiée, alors même que la compréhension de son déterminisme est essentielle pour évaluer les potentialités d’accommodation ou d’adaptation face au stress chimique. Nous nous attacherons notamment à distinguer la part de la polluosensibilité (et des traits morpho-anatomiques ou physiologiques associés) expliquée par la plasticité phénotypique des organismes, et celle liée aux différences entre génotypes. La variabilité génétique existant au niveau d’espèces modèles (e.g. Arabidopsis thaliana ; comparaison de mutants à la forme sauvage) sera par ailleurs mise à profit pour comprendre les mécanismes de transfert et les effets de certains nanomatériaux.
2) Quel est le rôle des effets directs et indirects des contaminants dans la structuration des communautés biologiques ?
Les impacts des contaminants à l’échelle des organismes / populations, même non létaux, sont susceptibles de modifier la diversité et l’abondance relative des espèces au sein des communautés. Au-delà de ses effets directs, la résultante de la contamination peut être fortement influencée par des effets indirects, liés à la modulation des interactions biotiques (compétition, prédation…) au sein des communautés.
Ces aspects seront notamment étudiés expérimentalement au travers des communautés de microorganismes (diatomées, bactéries…) qui, outre leur importance dans de nombreux processus écosystémiques, permettent des études à des échelles spatiales et temporelles compatibles avec des approches en laboratoire croisant de nombreux facteurs et impliquant un fort niveau de réplication. Ces approches seront replacées dans le cadre théorique de l’écologie des communautés, en nous appuyant notamment sur les modèles conceptuels intégrant les traits de réponse des organismes aux stress et aux perturbations.
Nous appréhenderons également l’impact des contaminants sur les communautés biologiques in situ, dans le cadre des chantiers d’étude in natura dans lesquels l’équipe ECI sera impliquée. Ces approches concerneront notamment les biofilms phototrophes en milieu aquatique, permettant ainsi de faire le lien avec les études expérimentales au laboratoire.
3) Quelles sont les conséquences fonctionnelles de la contamination chimique à l’échelle des écosystèmes ?
Les approches in natura permettent potentiellement d’appréhender l’impact d’une contamination sur les fonctions écosystémiques. Toutefois, il est nécessaire pour cela de disposer d’indicateurs de l’intégrité / altération fonctionnelle des écosystèmes.
La complexité de ces systèmes (et/ou des cocktails de contaminants en jeu) nécessite que ces indicateurs soient utilisables dans le cadre d’un aller-retour entre milieu naturel et conditions contrôlées. En outre, la prévision de l’impact de contaminants émergents (tels que les nanomatériaux), offrant par définition peu de possibilités d’étudier des situations de contamination in natura, implique le développement d’outils de bioindication et de compréhension des mécanismes qui soient utilisables au laboratoire ou en mésocosmse.
Nous construirons ces outils de manière à examiner les conséquences fonctionnelles des impacts sur les organismes (micro-organismes, plantes et métazoaires), en termes de processus d’importance écosystémique : les processus de bioturbation, de production primaire et de décomposition de la matière organique.
